These three amino acids form the core of the urea cycle, the hepatic pathway that converts toxic ammonia to urea, and are central to nitric oxide (NO) biosynthesis through their interconnected metabolic relationships:

L-Arginine is the direct substrate for nitric oxide synthase (NOS) enzymes, endothelial NOS (eNOS), neuronal NOS (nNOS), and inducible NOS (iNOS), which convert arginine to NO and citrulline. NO produced by eNOS in vascular endothelium activates soluble guanylyl cyclase (sGC) in smooth muscle cells, generating cGMP and driving vasodilation. This is the central mechanism of exercise-induced “pump”, increased blood flow and nutrient delivery to working muscle. L-arginine is also a substrate for arginase (producing ornithine + urea) and is involved in polyamine synthesis and creatine biosynthesis.

L-Citrulline is produced endogenously as a byproduct of NO synthesis. In the urea cycle, citrulline is converted back to arginine via argininosuccinate synthase and argininosuccinate lyase, effectively recycling the arginine pool and sustaining NO production longer than arginine supplementation alone. Research has consistently shown that L-citrulline raises plasma arginine levels more effectively than equivalent doses of L-arginine (due to citrulline escaping first-pass intestinal and hepatic arginase metabolism), though in an injectable context this advantage is less pronounced. Citrulline also directly reduces plasma ammonia during exercise, delaying fatigue in endurance models.

L-Ornithine is the urea cycle intermediate formed when arginase cleaves arginine. Ornithine is converted to citrulline in mitochondria by ornithine transcarbamylase (OTC), linking the urea and NO cycles. Ornithine supplementation has demonstrated ammonia-lowering effects in exercising subjects, reducing exercise-induced hyperammonemia and associated central fatigue. Ornithine also serves as a precursor to polyamines (putrescine, spermidine, spermine), essential regulators of cell growth, proliferation, and tissue repair.
